ABSTRACT

Objective To study the relationship between serum indoleamine 2 ,3-dioxygenase(IDO) and the level of regulatory T cells(Treg) in the patients with ulcerative colitis (UC) .Methods Seventy cases of UC treated in the Hainan Provincial People′s Hospital from January 2015 to February 2017 were selected as the observation group .In the same period ,70 healthy volunteers undergoing physical examination were selected as the control group .The levels of serum IDO and Treg were compared between the two groups ,the expression levels of serum IDO and Treg in of the patients with different disease activity periods and different severity de-grees of UC were observed ,and the correlation between the level of serum IDO and the Treg expression level in the patients with UC was analyzed .Results The serum IDO level in the observation group was significantly increased compared with that in the control group ,while the Treg expression level decreased which compared with the control group ,the difference was statistically significant (P<0 .05) .The serum IDO level in the pa-tients with activity period of the observation group was significantly increased compared with that in the pa-tients with remission period ,but the Treg expression level was decreased compared with that in the patients with remission period ,the difference was significant (P<0 .05) .The serum IDO and Treg expression levels had statistical difference among different disease degrees in the patients with active UC (P<0 .05) .The serum IDO level in the moderate and severe groups increased which compared with that in the mild group ,while the Treg expression level was significantly decreased compared with that in the mild group ,the difference was sig-nificant (P<0 .05) .The level of serum IDO in the severe group was significantly higher than that in the mod-erate group ,while the Treg expression level was significantly lower than that in the moderate group ,the difference was statistically significant ( P < 0 .05 ) .Conducting the correlation analysis according to the Spearman method found that there was a negative correlation between the serum IDO level and the Treg ex-pression level in the patients with UC (r= -0 .798 ,P=0 .000) .Conclusion The level of serum IDO in the pa-tients with UC is negatively correlated with the Treg expression level ,which jointly influence the occurrence and development of UC .

ABSTRACT

Objective To investigate the clinical characteristics and living conditions of hepatic diabetes (HD) patients and to analyze the risk factors of the prognosis of HD patient .Methods One hundred and sixty‐eight cases of cirrhosis cases were divided into liver cirrhosis group(n=74) ,the combined group cases(n=48)and combined HD type 2 diabetes (T2MD) group(n=46)se‐lected from January 2011 to December 2012 .Death was end point .The clinical features and prognosis of patients were compared . The risk factors for liver cirrhosis were analyzed by the simple factor analysis and Logistic regression analysis .Results The pa‐tients with history of diabetes in T2MD group were more than those in the HD group and the cirrhosis group (P0 .05) .The total bilirubin (TBil) of HD group was higher than those of the T2MD group and liver cirrhosis group(P<0 .05) .The levels of fasting glucose levels(FPG) ,insulin resistance index (HOMA‐IR) ,total cholesterol (TG) ,C‐peptide of liver cirrhosis group were lower than HD group and T2MD group (P<0 .05) .The levels of FPG ,C peptide of HD group were higher than that of T2MD group (P<0 .05) .The levels of TG of T2MD group were higher than that of HD group (P<0 .05) .Logistic multivariate analysis showed that age ,alcohol consump‐tion ,diabetes ,Child‐Pugh score were independent risk factors for death in patients with liver cirrhosis .Conclusion The morbidity and mortality of HD patients may be higher than patients with liver cirrhosis ,and HD could increase the risk of death in patients with liver cirrhosis .

ABSTRACT

Objective To observe the mRNA expressions of ornithine decarboxylase(ODC), endostatin and microvessel density (MVD) in esophageal squamous cell carcinoma (ESCC) and their correlations, and to explore the mechanisms of ODC in facilitating angiogenesis, its correlation with invasion a nd metastasis of the carcinoma. Methods Semi-quantitative RT-PCR method was used to detect the mRNA expressions of ODC and endostatin in 41 paired surgically resected specimens of ESCC (T)and their adjacent tissue(N), a nd the T/N ratio was calculated.Immunohistochemistry was used to detect the MVD by anti-CD 34 monoclonal antibody in ESCC tiss ue and corresponding adjacent tissue. Results Of 41 cases, the T/N ratio of ODC mRNA was greater than 1.0 in 39 cases (95.1%) and the T/N ratio of MVD was greater than 1.0 in 4 0 cases ( 97.6%). The mRNA expressions of ODC and endostatin, and MVD in esophageal specimens we re significantly correlated with tumor sizes, lymph node metastasis and adventit ia invasion. The mRNA expressions of ODC and endostatin were correlated with differentiation status of tumor. T/N value of ODC mRNA was positively correlated with T/N value of MVD, and nega tively correlated with the T/N value of endostatin mRNA. While the T/N value of endos tatin mRNA was negatively correlated with the T/N value of MVD. Conclusion ODC is closely related to tumor angiogenesis, invasion and metastasis of ESCC. It is possible that ODC overexpr ession may promote tumor angiogenesis by suppressing endostatin expression, which may be o ne of the mechanisms that ODC facilitates invasion and metastasis of ESCC.
